چکیده

The effect of excess of cadmium (Cd) on chlorophyll fluorescence of barley seedlings was studied. The leaves were dark-adapted and then illuminated at 10 different light intensities within the range of 70–1600 μmol.m–2.s–1 photosynthetically active radiation (PAR). It was found that Cd decreases the chlorophyll fluorescence parameters in dark-adapted leaves: ground (Fo), maximal (Fm) and variable (Fv) fluorescence. However the Fv/Fm ratio was slightly influenced. At light intensities below 400 μmol.m–2.s–1 PAR no significant differences between values of photochemical quenching (qP), non-photochemical quenching (qN) and yield of photochemical energy conversion (Y) of control and Cd-treated plants were found. At light intensities about 500 μmol.m–2.s–1 and higher significant decrease in qP, concomitant increase in qN as well as a reduction in Y were observed. In Cd-treated plants the maximum of apparent electron transport rate (ETR) were reached earlier than in control plants and under lower PAR intensities, indicating that the energy consumption by Calvin’s cycle reactions is reduced. It is speculated that excess of Cd in barley plants causes a down regulation of PSII to avoid over-reduction of primary electron acceptor QA and to reduce the load on the electron transport chain.

Salicylic acid alleviates the cadmium toxicity in barley seedlings.

Salicylic acid (SA) plays a key role in plant disease resistance and hypersensitive cell death but is also implicated in hardening responses to abiotic stressors. Cadmium (Cd) exposure increased the free SA contents of barley (Hordeum vulgare) roots by a factor of about 2. Cultivation of dry barley caryopses presoaked in SA-containing solution for only 6 h or single transient addition of SA at ...

Photosystem II of barley seedlings under cadmium and lead stress

Chlorophyll a fluorescence measurements were carried out on two barley (Hordeum vulgare L.) cultivars Arabi Abiad and Arabi Aswad at 8 and 14 days after emergence to identify their early tolerance mechanism for heavy metals (25 and 50μM of cadmium and lead). Transient fluorescence curves (OJIP curves) and energy flux models showed different specific reactions of photosystem II (PSII) of each cu...

Regulation of synthesis of the photosystem I reaction center

The in vivo biosynthesis of the P700 chlorophyll a-apoprotein was examined to determine whether this process is light regulated and to determine its relationship to chlorophyll accumulation during light-induced chloroplast development in barley (Hordeum vulgare L.). Rabbit antibodies to the 58,000-62,000-mol-wt apoprotein were used to measure relative synthesis rates by immunoprecipitation of i...

Physiological responses of resistant and susceptible barley, Hordeum vulgare to the Russian wheat aphid, Diurpahis noxia (Mordvilko)

Knowledge of the physiological responses of barley, Hordeum vulgare L., to the Russian wheat aphid, Diuraphis noxia (Mordvilko) (Hemiptera: Aphididae) is critical to understanding the defense response of barley to aphid injury and identifying resistance mechanisms. This study documented the impact of D. noxia feeding on resistant (‘Sidney’) and susceptible (‘Otis’) barley through chlorophyll fl...

Effect of Zn, Cd and Cr on growth, water status and chlorophyll content of barley plants (H. vulgare L.)

To evaluate the potential of barley for the phytoremediation of soils contaminated by metals, we conducted a growth chamber experiment with plants exposed to various concentrations of Zn, Cd and Cr. Growth parameters, chlorophyll content and chlorophyll fluorescence were measured at 15 and 29 days after treatment application, and the metal concentration in the aerial part of the plant, the root...
